Labeled antibodies are needed for applications such as WB, ELISA, immunofluorescence, immunohistochemistry (IHC), flow cytometry, screening of paired antibodies, and other experiments. Biologics International Corp (BIC) provides both antibody preparation services and professional antibody labeling services. Labeling molecules include fluorescein isothiocyanate (FITC), horseradish peroxidase (HRP), and biotin, which are suitable for the labeling of monoclonal, polyclonal,and recombinant antibodies. The labeled antibody could be used to localize and perform qualitative and quantitative analysis of the corresponding antigen.

Antibody Labeling

  • FITC labeling: FITC is used as a fluorophore. It has a maximum absorption wavelength of 490–495 nm and a maximum emission wavelength of 520–530 nm and shows yellowish green fluorescence. Under alkaline conditions, the lysine R-amino of the antibody can covalently bond to the thiocarbmide of FITC to form the antibody-FITC fluorescent complex. Tracing and qualitative/quantitative analysis are performed on the corresponding antigens with the use of fluorescence microscopy and flow cytometry.
  • HRP labeling: The carbohydrates on the surface of HRP are oxidized to aldehydes then covalently bound to the amino termini of antibodies to form an HRP-antibody complex. The color of HRP-labeled antibodies changes in the presence of specific enzymes and substrates. Qualitative/quantitative analysis of the antigens is based on color changes and HRP labeled technology is widely used in the analysis of WB, ELISA, IHC, etc.
  • Biotin labeling: In Vitro Biotin Labeling can biotinylate Avi tags or several amino groups of antibodies. Biotinylated antibodies can specifically bind to avidin which can be used to purification, IHC and ELISA assays.
