The family of Fibroblast Growth Factors (FGFs) consists of 23 proteins that regulate cell proliferation, migration, differentiation and survival. Fibroblast growth factor-23 (FGF-23) is a bone-derived phosphatonin that regulates phosphorus and vitamin D metabolism. FGF-23 is produced by osteocytes in bone and regulates phosphate homeostasis via signalling through its receptors (mainly FGFR1) in the presence of Klotho, its cofactor in the kidney and parathyroid glands. FGF-23 was originally identified as a gene mutated in patients with autosomal dominant hypophosphatemic rickets (ADHR). Nowadays, FGF-23 is elevated in patients with chronic kidney disease (CKD) and is strongly associated with mortality and cardiovascular disease. To exert its primary physiologic functions, FGF-23 binds to FGF receptor (FGFR)-klotho complexes in the kidney to stimulate urinary phosphate excretion and inhibit renal production of 1,25-dihydroxyvitamin D. Furthermore, FGF-23 also negatively regulates parathyroid hormone (PTH) secretion.

Formulation | Sterile Filtered white lyophilized powder. The FGF-23 protein (0.5mg/ml) was lyophilized from a 0.2µm filtered concentrated solution in PBS, pH 7.4. |
Stability | The lyophilized form of the protein is stable at room temperature for 3 weeks and at -20°C for at least 2 years from the date of receipt. Upon reconstitution, the protein should be stored at 4°C for no more than 7 days. The protein is stable at -20°C for six months. For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended. Please prevent freeze-thaw cycles. |
Biological Activity | The biological activity of FGF-23 was measured in a cell proliferation assay using NIH/3T3 mouse embryonic fibroblasts. The ED50 for this effect is typically 0.05-0.5µg/ml in the presence of 5µg/ml of Recombinant Mouse Klotho and 10 µg/ml of heparin. |
Solubility | It is recommended to reconstitute the lyophilized FGF-23 in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions. |
